Horne embarked on his comedic career and formed the comedy duo Mat and MacKinnon, making their debut at the Edinburgh Fringe in 2000. His talent caught the attention of Catherine Tate during the festival, leading to an invitation to join the BBC TV program, The Catherine Tate Show. Horne continued his involvement in several Catherine Tate’s Nan specials, reprising his role as Jamie in two festive episodes of Catherine Tate’s Nan in 2015.

In 2003, he took on a role in Channel 4’s 20 Things to do Before You’re 30, and the following year, he portrayed Ben, an atheist RE teacher, in the Channel 4 series Teachers. Horne also made appearances in the first series of ITV’s Doc Martin. Further, he starred in BBC sitcom Roman’s Empire in 2007 and participated in comedy sketches on ITV1’s The Kylie Show alongside Kylie Minogue. Notably, from 2007 to 2010, he played the main character Gavin Shipman in the double-BAFTA-winning series Gavin & Stacey, which received widespread critical acclaim and included a Christmas special. He reprised his role for the Christmas special in 2019.

Horne ventured into film, starring in Lesbian Vampire Killers with James Corden, and made his stage debut in a 2009 revival of Entertaining Mr Sloane. He experienced a health incident during a West End performance but recovered. In 2009, he co-presented the BRIT Awards with James Corden and Kylie Minogue.

In 2010, Horne appeared in Miss Marple’s episode “The Secret of Chimneys” and played Jon Moss in the BBC TV docudrama Worried About the Boy, depicting the teenage years of Boy George. He toured British theaters with Catherine Tate in The Catherine Tate Show Live and portrayed a young Mr. Grace in Are You Being Served?. He also played Private Walker in Dad’s Army: The Missing Episodes in 2019.

A feature film adaptation of Catherine Tate’s Nan, in which Horne was involved, was initially scheduled for release in June 2020 but was postponed due to the COVID-19 pandemic. It eventually premiered on March 18, 2022. In May 2022, Horne appeared in the BBC dark comedy series Inside No. 9, in the episode titled “Love Is A Stranger.”
