Ackles delved into an acting career in 1996, focusing on guest roles in series such as Mr. Rhodes, Sweet Valley High, and Cybill. Subsequently, he joined the cast of the NBC soap opera Days of Our Lives in 1997, portraying Eric Brady. Ackles garnered recognition, winning a 1998 Soap Opera Digest Award for Best Male Newcomer and receiving three Daytime Emmy Award nominations for Outstanding Younger Actor in a Drama Series in 1998, 1999, and 2000. In 2000, Ackles left Days of Our Lives and ventured into the mini-series Blonde, dedicated to Marilyn Monroe’s life.

Despite auditioning for the role of young Clark Kent on Smallville, he landed a guest role on Dark Angel in 2001, returning as a series regular in the second season. Following this, he worked on various projects, including Dawson’s Creek, the unaired series Still Life, and the 2004 film The Plight of Clownana. Ackles also declined a role in Tru Calling, leading to the character being renamed “Jensen.” Returning to Smallville in 2004, Ackles played Jason Teague, the assistant football coach. His involvement extended to the 2005 film Devour. Notably, in 2005, he joined the cast of Supernatural, a horror/action series on the WB, where he portrayed Dean Winchester. The show concluded after a remarkable fifteen-season run, establishing it as the longest-running North American sci-fi series.

In 2007, Ackles embraced the role of Priestly in the independent comedy Ten Inch Hero. He received acclaim for his comedic timing, and the film was released on DVD in 2009. Additionally, Ackles made his professional theatre debut in A Few Good Men in 2007. He expanded his filmography with the 2009 remake of My Bloody Valentine 3D and voiced Red Hood in the animated film Batman: Under The Red Hood in 2010.

Venturing into the realm of video games, Ackles voiced a character in Tron: Evolution in 2010. Beyond acting, he pursued a music career, releasing the song “Sounds of Someday” and forming a Radio Company with Steve Carlson. Their debut album, Vol. 1, was released in 2019, followed by Vol. 2 in 2021 and Keep on Ramblin in 2023. Ackles joined The Boys in its third season in 2020, portraying Ben / B.C.L. RED / Soldier Boy. He later played a version of Soldier Boy in the Gen V spin-off episode “Jumanji.” In collaboration with his wife, Ackles established Chaos Machine Productions, developing projects with Warner Bros. Television. Notably, they developed a Supernatural prequel spinoff for The CW, titled The Winchesters, which was picked up to series in 2022.

In October 2021, Ackles took on the role of U.S. Marshal Wood Helm in the Western feature Rust, alongside Alec Baldwin. Tragically, production was temporarily halted following the accidental death of cinematographer Halyna Hutchins. In May 2022, Ackles guest-starred in the ABC drama Big Sky’s second-season finale, “Catch a Few Fish,” as temporary sheriff Beau Arlen. The role was set to become a series regular for the show’s third season, which was ordered shortly after the episode aired.
